2D-DIGE and DALPC analysis of receptor tyrosine kinase signaling in human cancer cells

Abstract

The 2D difference gel electrophoresis (2D-DIGE) and DALPC analysis of receptor tyrosine kinase signaling in human cancer cells were studied. The DALPC technology was very sensitive in identifying proteins that were immunopurified with the receptors. The first dimensional isoelectric focusing was carried out using 24 cm immobilized pH gradient strips of varying pH gradients that were passively rehydrated in the presence of the mixed, pre-labeled samples. Second dimension MW separation was carried out using 12% acrylamide gels. A Sypro Ruby post-stain was used to image all of the proteins in the gel to ensure accurate robotic protein excision.
