Effect of transforming growth factor-β2 on expression of connective tissue growth factor mRNA and protein in cultured bovine lens epithelial cells

Abstract

• Aim: To observe the effects of transforming growth factor-β 2 (TGF-β2) on the mRNA and protein expression of connective tissue growth factor (CTGF) in bovine lens epithelial cells (LEC). • Methods: Cultured 2-3 passage bovine LEC in vitro were divided into control group and experimental group treated with 1,5,10μ g/L TGF-β2 for 24 hours respectively, The mRNA and protein of CTGF in bovine LEC were examined respectively by semiquantitative RT-PCR and Western blot, and the expression levels of CTGF were determined relative to that of β-actin. • Results: In comparison with control bovine LEC, 1μg/L -TGF-β2 did not affect the values of CTGF/μ-actin both in RT-PCR and Western blot (P > 0. 05). However, 5, 10μ g/L TGF-β2 significantly up-regulated the values of CTGF/ β-actin in the examinations (P< 0.01). • Conclusion: The data suggested that TGF-β2 could promote the expression of CTGF at mRNA and protein level obviously in bovine LEC. The role of CTGF in LEC transdifferentiation induced by TGF-β2 in pathogenesis of posterior capsule opacification is worthy of further investigation.
