Abstract
Antibodies to human alphafetoprotein were produced by hybridization of NS1/1 mouse myeloma cells with spleen cells from mice immunized with AFP. Four cell lines producing anti-AFP were established by cloning of cultures positive for anti-AFP from 3 separate hybridizations. These antibodies were all of IgG1 (k) type. Highly sensitive radioimmunoassays were established using these monoclonal anti-AFPs. These assays gave the same quantitative values for AFP in samples of serum and amniotic fluid as assays using conventional antisera. Our results demonstrate that monoclonal antibodies with promising performance in radioimmuno-assay can be obtained. Such antibodies may provide useful reagents in clinical assays and should also become useful tools in delineating the antigenic structure of the AFP molecule.
