The frequency and activity of single helper T cells

Abstract

The activities of T cell populations able to cooperate in the humoral immune response to TNP coupled to the carriers sheep red blood cells (SRBC) or keyhole limpet hemocyanin (KLH) were investigated. Poisson analyses were used in which small numbers of carrier primed spleen cells were added to cultures containing an excess of B cells and antigen. It was demonstrated that the activity of a single helper T cell could be detected and that its frequency in a given spleen cell suspension could be calculated as well as its relative activity in stimulating the humoral immune response.
