Evaluation of electron capture dissociation for D atom localization at the peptide and protein levels after solution phase exchange

Abstract

The electron capture dissociation (ECD) for localization of deuterium atom at peptide levels was investigated after solution phase exchange. THe deuterated samples were incubated for 4 hours at 75° C which was then cooled to sampling and deuterated protein was digested with pepsin. The digested protein was rapidly desalted with peptide trap and eluted with transfer line which was cooled by modified refrigerator. The deuterated experiments show back exchange of 40% for both ten second deuteration and maximally deuterated samples.
