Lanthanide-based dual imaging probes have great potential, but there are several obstacles that must be overcome before they can be translated into practical biomedical applications. High in vivo stability is necessary to minimize toxicity. We ad- dressed this problem by using a ligand system based on DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid), as these chelators form kinetically inert complexes. It is also desirable to shift the excitation wavelength above 600 nm to minimize absorption and scattering by biological tissue. This could be achieved by modifying the tropone chromophore.
